Induced breeding of cellulase producing strain and optimization of fermentation conditions

Abstract: The cellulase production strain Trichoderma viride T2 was treated with UV and EMS. The high cellulose production mutant T211 was obtained and its CMCase and FPA yields were 19.17 IU/mL and 1.94 IU/mL, respectively. The effects of compositions of culture medium and cultural conditions on cellulase production were investigated. The optimal culture media are: avicel 10 g/L, wheat bran 10 g/L, peptone 0.4 g/L, urea 0.4 g/L, (NH4)2SO4 2.0 g/L, tween-80 2.0 mL/L, and the others are same as Mandel’s medium. The optimal cultural conditions including incubating temperature, load of shake flask, rotation rate and amount of inoculation are: 27~28℃, 50 mL/250 mL,180 r/ min and 4%~6%, respectively. Under this condition, the CMCase and FPA yields reached 25 IU/mL and 2.5 IU/mL, respectively.
