Study on methods for isolation of nitrobacteria in aerobic sludge

Abstract: In this study, agar plate, agarose plate, washed agar plate and silicagel plate were used to isolate and culture nitrobacteria from aerobic activated sludge of municipal sewage plant. The results showed that, when culture conditions were the same, there were non-target bacteria growing both on the agar and washed agar plate, while that did not happen on the agarose plate, it was cumbersome to make silica gel plate, and dehydration and cleft appeared in the late cultures. The nitrobacteria was successfully separated out from agarose plate, which was identified by microscope observation and NorB gene detection, so agarose plate could be used for isolating and culturing the autotrophic nitrifying bacteria which has a long generation time.
